Abstract: Objective: To investigate the effection of IGF-域and EGF on the development of ICR mouse
preimplantation embryos in the CZB culture medium with glucose and insulin in vitro. Methods: 淤Experiment1:
One-cell ICR mouse embryos were cultured in the CZB medium with glucose and insulin supplemented with IGF-
域(0, 0.1, 1, 10 and 100 滋g/L), and a separate control group of CZB medium was established .于Experiment 2:
One-cell ICR mouse embryos were cultured in the CZB medium with glucose, insulin and IGF-域(0.1 滋g/L)
supplemented with EGF (0, 0.1, 1, 10 and 100 滋g/L), and a separate control group of CZB medium was
established. Blastocyst rates, hatched blastocyst rates and the cell numbers of blastocysts were counted. Results:
①Experiment1: IGF-Ⅱ(0 μg/L)increased the rate of blastocyst, hatched blastocyst and the total cell numbers of
blastocyst compared with control group(P<0.05). IGF-Ⅱat a dose of 100 μg/L decreased the rate of blastocyst,
hatched blastocyst and the total cell numbers of blastocyst compared with the other groups(P<0.05). IGF-Ⅱ at a
dose of 0.1 μg/L greatly increased the total cell numbers of blastocyst(P<0.05). ②Experiment 2: EGF(0 μg/L)
increased the rate of blastocyst, hatched blastocyst and the total cell numbers of blastocyst compared with control
group(P<0.05). EGF at doses of 0.1, 1 and 10 μg/L increased the rate of blastocyst and hatched blastocyst
compared with EGF at doses of 0 and 100 μg/L(P<0.05),and EGF at a dose of 0.1 μg/L had the highest rate of
hatched blastocyst(P<0.05). EGF at a dose of 0.1 μg/L increased the total cell numbers of blastocyst compared
with EGF at a dose of 0μg/L(P<0.05). EGF at a dose of 100 μg/L had the lowest rate of the total cell numbers
of blastocyst(P<0.05). Conclusions:淤IGF-域渊1, 10 滋g/L冤, glucose and insulin showed no significant effect of
suppression or promotion among. 于EGF 渊1, 10 滋g/L冤in CZB medium with glucose, insulin and IGF-域could
involves in the regulationof differentiation of embryonic cells but no proliferation. 盂IGF-域and EGF at a dose of
0.1 滋g/L is best for cell culture concentration. 榆IGF-域and EGF at a dose of 100 滋g/L have inhibition effect on
embryonic cells.
