前列腺素E2促进RL95-2细胞中整合素αv、β3的表达

摘要: 目的:探讨外源性前列腺素E2(PGE2)通过影响整合素αv、β3对胚胎黏附率的改变及可能的机制。方法:使用实时定量聚合酶链反应(real-time PCR)、蛋白质印迹(Western blotting)检测外源性给予PGE2后,RL95-2细胞的整合素αv、β3 mRNA及蛋白表达水平的变化。利用BeWo滋养层细胞和RL95-2上皮细胞株建立子宫内膜-滋养细胞相互作用的体外模型,观察BeWo细胞球在经过不同浓度PGE2预处理的RL95-2上皮细胞单层的黏附效率。结果:200 nmol/L PGE2预处理RL95-2细胞单层后促使整合素αv的mRNA及蛋白表达量比0 nmol/L组增加(均P<0.05);100 nmol/L PGE2预处理RL95-2细胞单层后促使整合素β3的mRNA及蛋白表达量比0 nmol/L组增加(均P<0.05)。200 nmol/L PGE2预处理后BeWo细胞球黏附到RL95-2细胞单层的效率明显提高。结论:适当浓度的PGE2可上调整合素αv、β3的表达,促进胚胎黏附效率。提供了在植入期间促进整合素αv、β3表达的进一步证据,这将有助于阐明通过改善子宫内膜容受性促进胚胎黏附的分子机制。

Abstract: Objective:To explore the effect of exogenous prostaglandin E2 on the embryonic adhesion rate by integrin αv and β3, as well as the potential mechanism. Methods:The expressions of integrin αv and β3 mRNA and protein in the cultured RL95-2 cells treated with exogenous PGE2 were detected by real-time PCR and Western blotting. BeWo trophoblast cells and RL95-2 endometrial epithelial cells were used to establish the in vitro model. The RL95-2 epithelial cell monolayer was pretreated by different concentrations of PGE2. The rate of the adhesion between BeWo cell and RL95-2 epithelial cell monolayer was compared. Results: PGE2 (200 nmol/L) significantly increased the expressions of integrin αv mRNA and protein (P<0.05), while PGE2 (100 nmol/L) also significantly increased the expressions of integrin β3 mRNA and protein in RL95-2 cells (P<0.05). The rate of the BeWo sphere cell adhered to the RL95-2 cell monolayer was significantly increased by the pretreatment with 200 nmol/L PGE2. Conclusions: PGE2 in appropriate concentration may increase the expressions of integrin αv and β3, and improve the embryonic adhesion. This study provides further evidence to promote the embryonic adhesion during implantation by increasing the expression of integrin αv and β3 and improving the endometrial receptivity.
