摘要: 糖原合成酶激酶3(glycogen synthase kinase-3,GSK-3)是一种广泛表达于各组织细胞的激酶,参与调节多种生物活动。在精子细胞中,其两种亚型(GSK-3α和GSK-3β)均有表达,参与调控精子在附睾中的成熟、女性生殖道中的获能和顶体反应等过程。GSK-3通过磷酸化隔膜蛋白、蛋白磷酸酶1γ2(PP1γ2)等靶蛋白影响精子获能与顶体反应。同时,作为参与了精子细胞中的Wnt信号通路、Toll样受体信号通路以及环磷酸腺苷/蛋白激酶A(cAMP/PKA)介导的蛋白质磷酸化通路等多条通路的激酶,GSK-3的活性也受到严格的控制。因此,研究GSK-3在精子成熟、获能和顶体反应中的作用对进一步阐明精子在附睾和女性生殖道中发生的各种生理过程和男性不育的发生机制具有重要的意义。

Abstract: Glycogen synthase kinase-3 (GSK-3), a kind of kinase expressed widely in different tissues and cells, is involved in the regulation of many biological processes. Two isoforms of GSK-3, GSK-3α and GSK-3β, are located in spermatozoon, suggesting that GSK-3 participate in the regulation of epididymal spermatozoon maturation, capacitation and acrosomal reaction in female reproductive tract. Through phosphorylating target proteins or enzymes such as septins4 and PP1γ2, GSK-3 could influence the sperm capacitation and acrosome reaction. As a vital kinase in many signaling pathways including Wnt signal pathway, Toll like receptor signaling pathway, cAMP/PKA mediated protein phosphorylation pathway and so on, GSK-3 is strictly regulated. Thus, the investigation about the role of GSK-3 in spermatozoon is important for elucidating the cause of male infertility as well as the physiological process of spermatozoon in the male and female reproductive tract.
