精原干细胞体外三维培养法的研究进展

摘要: 精原干细胞(spermatogonial stem cells,SSCs)位于睾丸曲细精管基底膜上,既能自我更新又能定向分化产生精子。SSCs这一特性在辅助生殖技术应用中发挥关键作用。多年来,从啮齿类动物SSCs到人类SSCs的体外增殖分化研究取得了极大的进步,这在一定程度上取决于体外培养法的不断改进和完善,目前SSCs体外培养已从最初的二维培养发展到各种类型的三维培养,如:软琼脂三维培养法、甲基纤维素三维培养法、纳米纤维支架三维培养法、脱细胞生物支架三维培养法。SSCs体外培养法的不断改进对揭示雄性生殖细胞发生机制、治疗男性不育症等具有重要意义。为了探索提高SSCs体外培养效率的技术方法,建立长期稳定的SSCs培养体系,本文主要综述了SSCs体外三维培养法的研究进展。

Abstract: Spermatogonial stem cells (SSCs) are located on the basement membrane of the testicular seminiferous tubules, which can not only renew themselves but also differentiate to produce sperm. The characteristics of SSCs are critical in assisted reproductive technologies. After years of research, the great progress has been made in in vitro proliferation and differentiation of SSCs from rodent SSCs to human SSCs, which depends on the continuous improvement of in vitro culture methods to a certain extent. At present, the in vitro culture of SSCs has developed from two-dimensional culture to three-dimensional culture, such as soft agar three-dimensional culture method, methylcellulose three-dimensional culture method, nanofiber scaffold three-dimensional culture method, and decellularized biological scaffold three-dimensional culture method. The continuous improvement of SSCs in vitro three-dimensional culture method is of great significance for revealing the mechanism of male germ cell generation and treating male infertility. In order to explore the technical methods for improving the in vitro culture efficiency of SSCs and establish the long-term and stable cell lines of SSCs, the research progress of in vitro three-dimensional culture methods of SSCs were reviewed in this paper.
