环状RNA在妊娠及妊娠相关疾病中的研究进展

摘要:

环状RNA(circular RNA,circRNA)作为一类重要的调控分子,可调控基因表达、介导信号通路,在妊娠这一复杂生理过程中发挥关键作用。研究表明,circRNA可通过竞争性内源RNA网络、表观遗传调控、蛋白互作等方式,参与胚胎发育、胎盘形成及母胎界面信号传导等核心生理过程,具体而言,circRNA参与调控滋养细胞功能、血管生成、糖脂代谢、免疫稳态以及蜕膜化进程等生物学过程。circRNA表达失调与子痫前期、妊娠期糖尿病、复发性流产、胎儿生长受限及妊娠期肝内胆汁淤积症等多种妊娠相关疾病的发病机制密切相关。未来有必要探索circRNA靶向干预策略,挖掘特异性circRNA作为妊娠相关疾病早期筛查标志物,可能有助于妊娠相关疾病的辅助诊疗。

Abstract:

Circular RNAs (circRNAs), as a group of crucial regulatory molecules, are capable of modulating gene expression and mediating signaling pathways, and exert vital functions in the sophisticated physiological process of pregnancy. Studies have demonstrated that circRNAs participate in the core physiological processes including embryonic development, placental formation and maternal-fetal interface signal transduction via multiple mechanisms such as competing endogenous RNA network, epigenetic regulation and protein-protein interaction. Specifically, circRNAs are involved in regulating trophoblast function, angiogenesis, glycolipid metabolism, immune homeostasis and decidualization. Dysregulated expressions of circRNAs are closely associated with the pathogenesis of multiple pregnancy-related diseases, including preeclampsia, gestational diabetes mellitus, recurrent spontaneous abortion, fetal growth restriction and intrahepatic cholestasis of pregnancy. It is necessary to explore the intervention strategies of circRNAs and identify specific circRNAs as early screening biomarkers for pregnancy-related diseases, which is helpful to facilitate the auxiliary diagnosis and treatment of pregnancy-related diseases.
