雌激素对去势大鼠SIRT1表达的影响

Effect of Estrogen on Expression of SIRT1 in Ovariectomized Rats

摘要: 目的:研究雌激素对去势大鼠大脑、心脏和肝脏等组织中沉默信息调节因子2相关酶1(SIRT1)表达的影响,以探讨其抗衰老作用。方法:选取3个月龄Sprague-Dawley(SD)雌性大鼠33只,手术切除大鼠双侧卵巢以建立大鼠去势模型,以部分模拟更年期的某些病理生理改变,分为实验组、去卵巢对照组和假手术对照组(每组各11只),实验组灌胃给予雌二醇(E2),去卵巢对照组和假手术对照组给予等量生理盐水,每天1次,共12周。满12周后腹主动脉采血,测定大鼠血卵泡刺激素(FSH)、E2水平,处死大鼠后取大脑、心脏、肝脏等组织。采用逆转录聚合酶链反应实验(RT-PCR)和酶联免疫吸附实验(ELISA)检测各组织SIRT1 mRNA及其蛋白水平,分析比较各组间SIRT1的表达水平。结果:大鼠灌胃12周后,去卵巢对照组FSH水平高于实验组和假手术对照组(P<0.001),E2水平低于实验组和假手术对照组(P<0.001)。在大鼠心脏、肝脏和脑组织中,实验组和假手术对照组SIRT1蛋白及mRNA水平均高于去卵巢对照组(P<0.05),实验组和假手术对照组SIRT1蛋白及mRNA水平差异无统计学意义(P>0.05)。在实验组和假手术对照组中,心脏SIRT1蛋白水平高于肝脏和脑组织(P<0.05),肝脏组织SIRT1蛋白量高于脑组织(P<0.05);而去卵巢对照组的心脏和肝脏组织SIRT1蛋白水平高于脑组织(P<0.05),但心脏和肝脏组织SIRT1蛋白水平差异无统计学意义(P>0.05)。3组心脏和脑组织SIRT1 mRNA水平均低于肝脏组织(P>0.05)。结论:雌激素可显著提高去势大鼠SIRT1蛋白及mRNA水平,进而发挥其抗衰老作用。

Abstract: Objective:To investigate the effects of estrogen replacement on the expression of silent information regulator 1(SIRT1) in those ovariectomized rats, so as to explore the anti-aging effect of estrogen. Methods:Thirty-three female Sprague-Dawley(SD) rats aged three months were used to develop the models by bilateral ovariectomy, which partly imitated the pathophysiological characteristics of menopause. There were three groups as follows: the treatment group(n=11) was treated with estradiol once a day for 12 weeks by intragastric administration in those ovariectomized rates; the sham-operated group(Sham,n=11) and the ovariectomized group(OVX,n=11) were treated with saline as controls. The levels of serum FSH and estradiol E2 was detected after 12 weeks′ treatment. The expressions of SIRT1 mRNA and protein in heart,liver and brain were analyzed by reverse transcriptional polymerase chain reaction(RT-PCR) and enzyme-linked immunosorbent assay(ELISA). Results:After 12 weeks intragastric administration in rats,the level of FSH in the OVX group was significantly increased compared with the treatment group and the OVX group(P<0.001),while the level of E2 was significantly decreased(P<0.001). In the heart,liver and brain tissues,the expression of SIRT1 protein and mRNA in the treatment group and the sham group were significantly higher than that in the OVX group(P<0.05),while there was not significantly difference between the treatment group and the sham group(P>0.05). In the treatment group and the sham group,the expression of SIRT1 protein in the heart tissue was significantly higher than that in the liver and brain tissues(P<0.05),while the expression of SIRT1 protein in the liver tissue was significantly higher than that in the brain tissue(P<0.05). However,the level of SIRT1 protein in the heart and liver tissues were higher than that in the brain tissues in the OVX group(P<0.05),while there was not significantly differences in the liver and brain tissues(P>0.05). In the three groups,the level of SIRT1 mRNA in the heart and brain tissues were lower than that in the liver tissue(P<0.05). Conclusions:Estrogen can significantly increase the expressions of SIRT1 protein and mRNA in the ovariectomized rats, suggesting its potential effect of anti-aging.
