摘要: 长链非编码RNA(long non-coding RNAs,lncRNAs)是一类长度超过200个核苷酸的不编码蛋白的RNA分子。近年来的研究发现,lncRNAs在转录水平、转录后水平以及表观遗传水平广泛参与调控基因表达以及染色质重塑等。简述lncRNAs的分类与3种主要的作用机制(诱导、引导和支架),总结近年来通过二代测序技术以及基因芯片技术鉴定到的在精子发生过程中表达的lncRNAs,重点介绍Mrhl、Tsx、HongrES2、Dmr、Tbca16、NLC1-C等lncRNAs在生殖细胞增殖、分化、成熟过程中的功能及作用机制,并对lncRNA的功能研究策略作出展望。

Abstract: Long non-coding RNAs (lncRNAs) are a group of RNA transcripts with longer than 200 nucleotides and without protein-coding capacity. Recently, it was revealed that the lncRNAs could be involved in the regulation of gene expression at the transcriptional, post-transcriptional and epigenetic levels, and chromatin remodeling. In this review, we summarized the classification of lncRNAs and three major mechanisms including decoys, guides and scaffolds. More and more lncRNAs related to spermatogenesis have been identificated by the next generation sequencing and microarray. The roles and potential mechanisms of lncRNAs in the proliferation, differentiation and maturation of germ cells were introduced, including those lncRNAs as Mrhl, Tsx, HongrES2, Dmr, Tbca16 and NLC1-C. Finally, the study strategy of lncRNAs was discussed.
